Role of Prokineticin in Epicardial Progenitor Cell Differentiation to Regenerate Heart
نویسنده
چکیده
Over the last decade, stem/progenitor-cell therapy has emerged as an innovative approach to provide cardiac repair and regeneration (Zimmermann, et al., 2006). Several stemand progenitor-cell types from autologous and allogeneic donors have been analyzed to find the most appropriate candidate. Although embryonic stem (ES) cells can differentiate into most cardiac cell types (Mummery, et al., 2002) , their clinical use is severely limited due to ethical concerns and immunogenic and teratogenic side effects (Blum and Benvenisty, 2008). Adult bone marrow-derived stem cells avoid the ethical and clinical issues associated with ES cells (Bianco, et al., 2001). However, animal studies have demonstrated a variable degree of cardiomyogenesis, and improvement in heart function by bone marrow-derived stem cells (Murry, et al., 2004). Thus, the utility of adult bone marrow-derived stem cells is hampered by their limited population size and restricted potential for cardiovascular differentiation (Assmus, et al., 2010).

OBJECTIVE Identification of novel factors that contribute to myocardial repair and collateral vessel growth hold promise for treatment of heart diseases. We have shown that transient prokineticin receptor-1 (PKR1) gene transfer protects the heart against myocardial infarction in a mouse model. Here, we investigated the role of excessive PKR1 signaling in heart. METHODS AND RESULTS Transgenic ...

Objective(s): Throughout evolution, mammalians have increasingly lost their ability to regenerate structures however rabbits are exceptional since they develop a blastema in their ear wound for regeneration purposes. Blastema consists of a group of undifferentiated cells capable of dividing and differentiating into the ear tissue. The objective of the present study is to isolate, culture expa...

OBJECTIVE Prokineticins are potent angiogenic hormones that use 2 receptors, prokineticin receptor-1 (PKR1) and PKR2, with important therapeutic use in anticancer therapy. Observations of cardiac and renal toxicity in cancer patients treated with antiangiogenic compounds led us to explore how PKR1 signaling functioned in heart and kidney in vivo. METHODS AND RESULTS We generated mice with a c...
متن کاملProkineticin receptor-1 signaling promotes Epicardial to Mesenchymal Transition during heart development
The epicardium plays an essential role in coronary artery formation and myocardial development. However, signals controlling the developing epicardium and epicardial-mesenchymal transition (EMT) in the normal and diseased adult heart are studied less rigorously. Here we investigated the role of angiogenic hormone, prokineticin-2 and its receptor PKR1 in the epicardium of developing and adult he...
متن کاملAre Stem Cells the next Therapeutic Tool for Heart Repair?
Cardiovascular disease remains the leading cause of morbidity and mortality in the United States and Europe. In recent years, the understanding that regenerative processes exist at the level of the myocardium, has placed stem cell research at center stage in cardiology. A stem cell is a cell that has the ability to divide (self replicate) for indefinite periods often throughout the life of the ...
